摘要:
目的:研究松潘乌头总碱对小鼠的半数致死量(LD50)，为药理实验和临床安全合理用药提供依据。方法:将松潘乌头粉碎后，用碳酸钠溶液浸润，经乙醇超声提取，硫酸溶解，氨水调pH，用乙醚：氯仿(3：1)混合溶液萃取，得松潘乌头总碱。根据预实验结果将60只小鼠分为6组，按照200、160、128、102.4、81.9、65.5 mg/kg的剂量一次性灌服小鼠后观察24小时内情况。记录小鼠中毒症状及死亡情况，观察松潘乌头总碱的急性毒性。结果:松潘乌头总碱经口服给药小鼠的LD50为119.78 mg/kg，其95%可信区间为103.86～139.28 mg/kg。结论:松潘乌头总碱有中等毒性，其半数致死量为119.78 mg/kg。
Abstract:
Objective: To provide the reference for pharmacological experiment and rational medication in clinic by study lethal median dose of total alkaloids of SongPan WuTou(Aconitum sungpanense Hand.-Mazz.). Methods: Total alkaloids of SongPan WuTou were obtained by smashing SongPan WuTou, soaking it in sodium carbonate solution, processing by ethanol ultrasonic extraction, dissolving in sulphuric acid, adjusting the pH of the solution, extracting by the mixture of aether and chloroform. According to the results of preliminary experiment, sixty mice were randomized into six groups, the conditions within 24 hours were observed after the mice received gavage of total alkaloids of SongPan WuTou according to the doses: 200, 160, 128, 102.4, 81.9 and 65.5 mg/kg. Poisoning symptoms and the death of the mice were recorded to observe acute toxicity of total alkaloids of SongPan WuTou. Results: LD50 of total alkaloids of SongPan WuTou was 119.78mg/kg, its 95% credibility intervals were from 103.86to 139.28 mg/kg. Conclusion: Total alkaloids of SongPan WuTou show moderate toxicity, and its LD50 is 119.78 mg/kg .
