摘要:
目的探讨阳虚体质膝骨性关节炎(knee osteoarthritis,KOA)患者雌二醇(estradiol,E2)水平对Notch1、Notch2信号通路的影响及相关性研究。 方法选取60例KOA患者,根据体质分型分为阳虚体质组与非阳虚体质组,每组30例。空腹采集外周静脉血,采用电化学发光法检测E2浓度;采用q-PCR与Western Blot实验方法检测血浆Notch1、Notch2的miRNA与蛋白表达量;E2浓度与Notch1、Notch2基因和蛋白相对表达量的相关性采用Spearman秩相关进行分析。 结果血清E2检测结果显示阳虚体质组E2浓度低于非阳虚体质组(P<0.05);Notch1、Notch2的mRNA和蛋白相对表达量组间比较,差异均有统计学意义(P<0.05)。阳虚体质组Notch1、Notch2 mRNA及蛋白相对表达量均高于非阳虚体质组(P<0.001)。阳虚体质组E2水平与Notch1、Notch2 mRNA和蛋白表均存在负相关性(P<0.001)。 结论阳虚体质KOA患者中E2浓度与Notch1、Notch2基因和蛋白呈明显的负相关,推测阳虚体质患者E2水平下降,可能是促使Notch1、Notch2基因和蛋白的表达升高,各种炎性因子上升,加快KOA发生发展的潜在原因,将E2-Notch信号通路与中医体质研究相结合,寻找KOA潜在治疗靶点,可能对KOA的治疗具有一定的科学价值。
Abstract:
ObjectiveTo discuss the influence of the levels of estradiol (E2) in knee osteoarthritis (KOA) patients on Notch1 and Notch2 signaling pathway, and its correlational study. MethodsSixty KOA patients were chosen, and divided into Yang deficiency constitution group and non Yang deficiency constitution group in light of the constitutions, with 30 cases in each group. After collecting peripheral venous blood on an empty stomach, electrochemical luminescence (ECL) was adopted to detect the concentrations of E2, q-PCR and Western Blotting were applied to measure the expressions of miRNA and protein of plasma Notch1 and Notch2, the correlation between the concentrations of E2, and the relative expressions of Notch1 and Notch2 gene and protein was explored using Spearman's correlation. ResultsThe results of serum E2 showed that the concentrations of E2 in Yang deficiency constitution group were lower than these of non Yang deficiency constitution group (P<0.05); the difference had statistical meaning in the relative expressions of mRNA and protein of Notch1 and Notch2 (P<0.05). Yang deficiency constitution group was higher than non Yang deficiency constitution group in the relative expressions of mRNA and protein of Notch1 and Notch2 (P<0.001). The levels of E2 were negatively related to the relative expressions of mRNA and protein of Notch1 and Notch2 in Yang deficiency constitution group (P<0.001). ConclusionThe concentrations of E2 and the expressions of Notch1 and Notch2 gene and protein represent a significant negative correlation, speculating that the decrease of E2 in Yang deficiency constitution patients might be the potential reason of raising the expressions of Notch1 and Notch2 gene and protein, the rise of the inflammatory factors and accelerating the incidence and development of KOA, the survey of the potential therapeutic targets of KOA by combining E2-Notch signaling pathway with the study on TCM constitutions might possess certain scientific value for the treatment of KOA.
