摘要:
目的探讨麻杏石甘汤通过调控Toll样受体4(Toll-like receptor 4 polypeptide,TLR4)/核因子-κB(nuclear factor-kappa B,NF-κB)信号通路治疗慢性阻塞性肺疾病(chronic obstructive pulmonary disease,COPD)的机制。 方法将36只SD大鼠随机分成正常组、模型组、治疗组,每组12只。除正常组外,模型组和治疗组采用烟熏联合气管内滴入脂多糖的方法制备COPD大鼠模型。正常组及模型组正常饲养,治疗组予以麻杏石甘汤灌胃治疗,共干预4周。采用肺功能仪检测大鼠肺功能[吸气阻力(inspiratory resistance,Ri)、呼气阻力(respiratory resistance,Re)和肺组织顺应性(dynamic lung compliance,Cdyn)];采用HE染色观察各组肺组织形态;采用Western blot检测基质金属蛋白酶9(matrix metalloproteinases-9,MMP-9)、TLR4、NF-κB p65蛋白相对表达量;采用ELISA法检测炎症因子白细胞介素6(interleukin-6,IL-6)和白细胞介素18(interleukin-18,IL-18)的表达。 结果模型组和治疗组Ri、Re显著高于正常组(P<0.05),Cdyn显著低于正常组(P<0.05);治疗组Ri、Re显著低于模型组(P<0.05),Cdyn显著高于模型组(P<0.05)。HE染色显示,正常组肺组织结构正常;模型组肺组织中较多炎性粒子浸润,肺组织结构破坏严重;治疗组肺组织形态较模型组明显改善。Western blot显示,模型组和治疗组MMP-9、TLR4、NF-κB p65蛋白相对表达量显著高于正常组(P<0.05);治疗组MMP-9、TLR4、NF-κB p65蛋白相对表达量低于模型组(P<0.05)。ELISA结果显示,模型组和治疗组IL-6和IL-18含量显著高于正常组(P<0.05),治疗组IL-6和IL-18含量显著低于模型组(P<0.05)。 结论麻杏石甘汤通过抑制TLR4/NF-κB信号通路,减少炎性细胞浸润及MMP-9的表达,从而改善COPD症状。
Abstract:
ObjectiveTo discuss the mechanism of Maxing Shigan decoction in the treatment of COPD by adjusting TLR4/NF-κB signaling pathway. MethodsA total of 36 SD rats were randomized into the normal group, the model group and the treatment group, 12 rats in each group. Except for the normal group, COPD rat models were established by adopting smoking combined with intratracheal instillation of lipopolysaccharide in the model group and the treatment group. The normal group and the model group were raised by normal forage, and the treatment group accepted oral administration of Maxing Shigan decoction, altogether for four weeks of intervention. Spirometer was adopted to detect lung function (Ri, Re and Cdyn) of the rats; HE staining was used to observe lung histomorphology in different groups; Western blot to measure the relative expressions of MMP-9, TLR4 and NF-κB p65 protein; ELISA to detect the expressions of IL-6 and IL-18. ResultsRi and Re of the model group and the treatment group were higher than these of the normal group (P<0.05), Cdyn lower than that of the normal group remarkably (P<0.05); Ri and Re of the treatment group were lower than these of the model group (P<0.05), Cdyn higher than that of the model group significantly (P<0.05). HE staining displayed that lung structure of the normal group was normal; while more inflammatory particles were infiltrated into lung tissue in the model group, the damage to the lung structure was severe; morphological improvements of lung tissue of the treatment group were more noticeable than these of the model group. Western blot results showed that the relative expressions of MMP-9, TLR4 and NF-κB p65 protein of the model group and the treatment group were higher than these of the normal group apparently (P<0.05); the relative expressions of MMP-9, TLR4 and NF-κB p65 protein of the treatment group were lower than these of the model group (P<0.05). ELISA results showed that the model group and the treatment group were higher than the normal group in the contents of IL-6 and IL-18 (P<0.05), the treatment group was lower than the model group in the contents of IL-6 and IL-18 (P<0.05). ConclusionMaxing Shigan decoction could improve the symptoms of COPD by inhibiting TLR4/NF-κB signaling pathway, reducing inflammatory cell infiltration and the expressions of MMP-9.
